His father, when he was a baby, had picked up the baby son, Dio, of a dying man by the last name of Brando. Dio spent most of his time growing up bullying Jonathan, going so far as to throw Jonathan's pet dog in a furnace and stealing the first kiss from Jonathan's prim and proper girlfriend, Erina.
However Dio turned his attention to stealing Jonathan's status as the good son, pretending to be become Jonathan's best friend in the process. During that time Jonathan's father got sick and the medicine Dio brought did nothing as he got sicker.
Jonathan went to a dangerous town in to find out what the medicine was, being rescued by a man named Caesar Zepelli who taught him a new form of martial arts, and a world-wise man named Robert Edward O. Speedwagon, who became a trusted friend of the Joestar family and re-encountered his girlfriend from years ago who had been shamed by Dio. In that time he found out the medicine was actually an ancient Chinese poison.
In the meantime, Dio accidentally got blood on an ancient stone mask owned by the Joestar family for years, and tested it on a homeless man. The man became a ferocious vampire who was about to devour Dio just as the sun rose, disintegrating him.
Jonathan, Zepelli, Erina, and Speedwagon brought home evidence that Dio was poisoning his adoptive father, which Dio didn't deny, instead cutting himself to put blood on the mask and putting the mask on his face becoming a vampire. A fight ensued that set the Joestar home on fire, and everyone escaped but Jonathan's father, who died, and Dio who was crushed by a collapsing ceiling. It would seem that his vampire abilities couldn't regenerate him faster than the fire could destroy him. All was good.
Months later Jonathan and Erina, now his wife were on a ship celebrating their marriage when Dio's head appeared on the boat causing chaos. Jonathan stayed behind on the boat to finish off Dio while his wife left on a lifeboat and watched the boat sink.
