Ok, since no one else answered it...

The episode was the one that featured Barclay's Protomorphosis
Syndrome. Everyone on the ship was morphing into one
primitive life-form or another.

Except Data, who is not biological.
Even Spot changed-but her kittens did not, which gave Data
the clue he needed. He found an expectant mother on the
ship, and synthesized a cure by examining her amniotic fluid's
makeup and proceeding from there.

That was one of the episodes where Spot was female.
Spot was male in earlier seasons, and the technically-minded
Data called Spot "him" during those seasons.
